Ferritin as a reporter gene of in vivo stem cell tracking by 9.4-T cardiac MR in a rat model of myocardial infarction

چکیده
Methods We tested human ferritin heavy chain (hFTH) tagged with both myc and green fluorescence protein (GFP) as a reporter gene for in vivo tracking of stem cells by cardiac MR. Rat mesenchymal stem cells (rMSCs) were transduced with lentiviurs to overexpress hFTH. Myocardia infarction was induced by cryoinjury in rats, and the animals were immediately subjected to intramyocardial injection of 100 μl of 106 rMSCs (experiment group) or buffer solution (control group) in the border zone. In vivo cine and in vivo and ex vivo multiecho T2 weighted images were obtained by 9.4T cardiac MR.

Ferritin as a reporter gene for in vivo tracking of stem cells by 1.5-T cardiac MRI in a rat model of myocardial infarction.
The methods currently utilized to track stem cells by cardiac MRI are affected by important limitations, and new solutions are needed. We tested human ferritin heavy chain (hFTH) as a reporter gene for in vivo tracking of stem cells by cardiac MRI. Swine cardiac stem/progenitor cells were transduced with a lentiviral vector to overexpress hFTH and cultured to obtain cardiospheres (Cs). Myocardi...

متن کاملThe Effect of Eight Weeks of Intermittent Training with Quercetin Encapsulated Nanoparticles on JNK and ERK Apoptosis Regulators in the Rat Model of Myocardial Infarction
Introduction: Restoration of blood flow to the ischemic heart muscle causes infarction and myocyte death. Quercetin can be effective in treating heart disease due to its antioxidant effect, but its use is limited due to its low solubility in water. Quercetin restriction can be removed by embedding it in carrier nanoparticles. The present study was designed to determine the effect of quercetin e...
متن کاملCardiac CEST-MRI for tracking stem cell survival and determining the role of CXCL2
Background Because of the limited endogenous repair mechanisms that exist in the heart, exogenous stem cell therapies offer the potential to provide the building blocks for repair and/or evoke developmental pathways to recruit endogenous cells that can prevent adverse remodeling and promote healing. MR-labeling of stem cells with iron oxides has been used to track stem cells in the heart, but c...
متن کاملAre Stem Cells the next Therapeutic Tool for Heart Repair?
Cardiovascular disease remains the leading cause of morbidity and mortality in the United States and Europe. In recent years, the understanding that regenerative processes exist at the level of the myocardium, has placed stem cell research at center stage in cardiology. A stem cell is a cell that has the ability to divide (self replicate) for indefinite periods often throughout the life of the ...
